序言Human body consists of many organs where many cells work together to perform their functions. Although massivemolecular data are averages over ensembles of cells, the question is whether all the cells in an organ work in thesame way. In a cancer tissue, it has been pointed out that a small amount of cancer stem cells play important rolesto reconstruct a cancer tissue again even after destroying the cancer tissue by anti-cancer drugs. However, the de-tails have not been clarified because of the difficulty of analyzing very small number of cancer stem cells. It has beenalso pointed out that the heterogeneity of ES cells might be important for keeping their totipotency. For under-standing a real life system, we need new tools to analyze individual cells besides the averaged biological data. Weare at a new frontier where we understand functions of organs based on the information on individual cells usingnew tools.Seven years ago, we started IFPT to promote the interdisciplinary and international collaborations in the post ge-nome era. Beijing is the capital of China and has many beautiful places. I hope all the participants will relax and en-joy the meeting in Beijing and hopefully have chances for collaborations.
文摘There is increasing evidence that microRNAs are involved in the pathogenesis of ischemic diseases such as post-ischemic mouse hind limb, and myocardial ischemia resulted in infarction. MiR-210 expression was uregulated inresponse to hypoxia in vivo and in vitro and evidence showed that it strongly regulated human endothelial cellsinduced by hypoxia in vitro. In I/R model, the expression of miRNA-210 was increased at early time pointsfollowing I/R (1——3 days) relative to levels obtained in shams, miR-17~92 cluster is highly expressed in humanendothelial cells and that miR-92a, a component of this cluster, controls the growth of new blood vessels(angiogenesis). Lately, Angelika Bonauer et al. identified that overexpression of miR-92a in endothelial cellsblocked angiogenesis in vitro and in vivo. In mouse models of limb ischemia and myocardial infarction, systemicadministration of an antagomir designed to inhibit miR-92a led to enhanced blood vessel growth and functionalrecovery of damaged tissue. The mouse hind limb ischemic model to study the expression level of miR-92a inskeletal muscle changes in response to ischemic injury. Ischemic injury significantly increased miR-92a expression, with a maximal effect seen within 1 to 3 days after injury and then antagomir-92a can promote neovascularization inischemic limbs.Tubular regeneration is an important element of the initial recovery from AKI induced by I/R; however, thestructural recovery of the vasculature is impaired and this may be critical in the compromise of long-term functionfollowing I/R. Although both structural and functional alterations in microvascular cells have been described withinhours of kidney I/R injury, whether the mechanisms of microvascular cells loss are related with expression ofmicroRNAs have not been delineated. Studies have suggested that miR-210 and miRNA-92a, which controlledangiogenesis and functional recovery of ischemic tissues such as hind-limp and heart in mice, were up-regulated
